Longevity factor klotho enhances cognition in aged nonhuman primates
A single low peripheral dose of alpha-klotho enhanced cognition in aged rhesus monkeys, with benefit persisting well beyond the protein's circulating half-life. This is the strongest non-human primate evidence for klotho as a cognitive intervention and the basis of most current translational klotho programmes. It has not been replicated in humans, and no approved alpha-klotho product exists.

Key Findings
- 01
Single low-dose peripheral administration of alpha-klotho improved cognition in aged rhesus monkeys
- 02
Cognitive benefit outlasted the circulating half-life of the administered protein
- 03
Effect occurred at a dose not expected to cross the blood-brain barrier appreciably, implying an indirect peripherally initiated mechanism
- 04
Provides the strongest non-human primate rationale for klotho-directed cognitive therapeutics
Structured Methods
- Study Design
- Preclinical Study
- Sample Size
- Not reported
- Study Duration
- Not reported
- Methodology
- Peripheral administration of recombinant alpha-klotho to aged rhesus macaques with subsequent cognitive testing.
- Limitations
- Non-human primate study — no human data. Small cohort typical of primate work. Single-dose design; chronic dosing safety and durability untested. The mechanism by which a peripherally administered dose produces central cognitive effects is not established. No approved alpha-klotho product exists for any indication.
