    Therapeutic inhibition of telomeric DNA damage response rescues hematopoietic dysfunction driven by telomere shortening and aging

    Oppezzo and colleagues targeted telomeric noncoding RNAs with telomeric antisense oligonucleotides (tASO) to suppress the telomeric DNA damage response (tDDR) in hematopoietic organs. In telomerase-deficient and physiologically aged mice, tASO reduced senescence and inflammation, alleviated hematopoietic dysfunction and enhanced hematopoietic stem cell fitness. Ex vivo tASO treatment also improved the function of human HSCs from aged donors. The conceptual advance is decoupling the damage signal from the damage itself, making tDDR druggable without restoring telomerase.

    Limitations
    Mouse in vivo plus human ex vivo only. No in vivo human exposure, no safety or pharmacokinetic data in people, and no lifespan endpoint. Antisense oligonucleotide delivery to human marrow in vivo remains unsolved.
