Lifelong restriction of dietary valine has sex-specific benefits for health and lifespan in mice
Peer-reviewed Nature Aging study (Calubag et al., Lamming lab) showing that restricting the single branched-chain amino acid valine — rather than all three BCAAs — improves metabolic health in both sexes and extends median lifespan by 23% in male C57BL/6J mice. Frailty, cancer prevalence and senescent-cell burden fell in both sexes; lifespan extension was male-specific.

- Limitations
- Rodent study in a single inbred strain (C57BL/6J). Lifespan extension was male-only; females showed healthspan but not lifespan benefit, which constrains translational generalisation. No human data on valine-restricted formulations, and long-term low-valine intake in humans carries unstudied risks for muscle protein synthesis in older adults.
